1: What in the world would they do? Unless its something crazy, but one would need to know what the crazy thing is before you can plan for it,

2: RB + Killer: Would probably try and bait PR's into using their power on someone, block and murder, then let things progress from there,
2: Killer + Something else: Idk,

I doubt there are. Very much. 5v1v3? Very unlikely. Also, what would the the 3rd mafia role even be? A killer, a RB'er, and...?

I suppose it could be a substituye killer, bit what sense would that make in a step where there's suppossed to be just one Demon lord?

Well, we know there is a disciple, and we know they are able to reveal alignment just like the prophet, but it would be far too op to have two simultaneous prophets. Also, Jailbreak—who seems to be Disciple due to various hints, such as his roleplay on D1 and the fact that not once did he refute the constant statements that assumed he was Disciple—has not cleared or sussed anybody, meaning he likely wasnt able to perform investigations these last two nights.
